A study of the relationship of structure and activity of antimycin A in the induction of carotenoid synthesis in Mycobacterium marinum.

چکیده
Mycobacterium marinum produces carotenoids when exposed to light or when antimycin A is added. Structural characteristics of antiiycin A in relationship to its induction of carotenoid synthesis were determined in order to make comparisons with the characteristics required for its inhibition of the electron transport system. All four components of the antimycin A complex (AI, Az, AZ, and A4) were equally active. Studies with a number of derivatives and synthetic analogues showed that the substituted dilactone ring and the phenolic hydroxyl group, but not the N-formyl (or an Ncarbonyl) group, are indispensable for the induction activity of antimycin A. Since the N-formyl group (together with the phenolic hydroxyl group and the dilactone ring) is required for the inhibitory activity of antimycin A in the electron transport system, the possibility that antimycin A induces carotenogenesis by interfering in the electron transport system was, therefore, discounted. Additional evidence for the lack of involvement of the electron transport system in the induction of carotenogenesis was provided when it was found that the replacement of the dilactone ring with a long alkyl chain resulted in the total loss of induction activity but not of inhibitory activity. A hypothesis that antimycin A and light induce carotenogenesis by inducing protein synthesis has been evaluated.

The Impact of Overexpression of Sigma Factors on Morphological Changes, Growth Pattern, and Biofilm Formation in Mycobacterium marinum CCUG 20998
Introduction: Bacteria have at least one sigma factor (σ-factor) that transcribes the genes required for cell viability. Usually, transcription of σ-factors occurs and changes in response to a variety of environmental stresses. Expression of σ-factors is one of the strategies which is used in response to different stress conditions. This study was aimed to evaluate the effects...
متن کاملMycobacterium marinum as a cause of skin chronic granulomatous in the hand
Background: Mycobacterium marinum infection has a worldwide distribution and the organism occupies many aquatic environments. Infections in human consist of nodular cutaneous lesions that can progress to tenosynovitis, arthritis, and osteomyelitis. Case presentation: A 27-year-old man was presented with a history of swelling and exudative erythematous lesions and pustules of the right forearm. ...
متن کاملIdentification of cutaneous granuloma caused by Mycobacterium marinum using PCR method
Atypical Mycobacterium granulomatous skin infections are often accured by Mycobacterium marinum, M. ulcerans, M. fortuitum, and M. avium colonies. Skin infections probably originate from an environmental source such as contacting with aquatic animals, fish farming and swimming in the pools, and inoculate into skin through skin wounds, scratches, trauma, and surgery. The lesions appear as purple...
